Background of GEN1042-CD137-hIgG1

4-1BB (CD137) is a costimulatory TNFR expressed on immune cells, including T cells, natural killer cells and monocytes. While naïve T cells lack 4-1BB expression, the receptor is upregulated on T-cell receptor engagement. Crosslinking of 4-1BB activates the MAPK and NF-κB signaling pathways and enhances T-cell proliferation and cytokine production, enhances cytotoxic T-cell effector functions and prevents activation-induced cell death of T cells. GEN1042 (DuoBody®-CD40x4-1BB) is an investigational, novel, agonistic, bispecific antibody that combines targeting and conditional activation of CD40 and 4-1BB on immune cells for priming and (re-) activation of tumor-specific immune responses. GEN1042-CD137-hIgG1 is a monoclonal antibody that only targets CD137.
