Background of AMG305-CDH3-hIgG1

AMG 305 is a pioneering BiTE庐 (bispecific T cell engager) molecule that simultaneously binds to both P-cadherin (CDH3) and mesothelin (MSLN) to exert potent cytotoxic effects. Immunohistochemistry analysis reveals that CDH3 and MSLN are co-expressed in various types of solid tumors. However, in normal tissues, immunostaining demonstrates a lower occurrence of overlapping co-expression compared to the expression of either one of the targets alone. AMG 305 employs an avidity-based mechanism to activate T cells, enabling them to selectively eliminate CDH3+MSLN+ tumor cells while displaying limited activity against normal cells expressing only one of the target antigens.
