Background of Tarlatamab

Delta-like ligand 3 (DLL3) is an inhibitory Notch ligand that is highly expressed in SCLC and other neuroendocrine tumors but minimally expressed in normal tissues. It is therefore being explored as a potential therapeutic target in SCLC. Tarlatamab, a half-life extended bispecific T-cell engager (HLE BiTE) molecule, binds both DLL3 on cancer cells and CD3 on T cells leading to T-cell–mediated tumor lysis. Tarlatamab promotes tumor regression in preclinical models of SCLC.17 Tarlatamab is the first DLL3-targeted immune therapy to be evaluated clinically in SCLC.
